Is "what do you want?" a hostile response to a greeting? When you great someone and they respond with "what do you want?" would you see that as negative?

Tone plays a large role as well.
How much wood would a woodchuck chuck if a woodchuck could chuck wood?

Anyone who, in this day and age, is offended by "what do you want" as a greeting needs to grow a fucking pair and get the fuck over it. Seriously. You make a greeting, I respond pointedly telling you I don't have time do deal with any fucking timewasting bullshit, tell me what you want or get the fuck out. It's as simple as that. Anyone who disagrees has way too much time on his or her hands and needs to go do something productive.

It was an internet chat that spawned this question. Tone doesn't translate.

Also, Holy, take a deep breath. No one was offended.

This is about winning a disagreement with Spaniard.

Tone is crucial, but since you're on the internets you'd kind of have to guess the mood the other guy's in. If he follows it up with an xD or something then it's polite, but if the first thing he says after hello is "what do you want" and doesn't follow it up it's considered rude. Hail english.

What sort of tone can you use where saying "what do you want?" to someone saying hello to you is not considered rude?

Rude.  Worth getting upset about, no.  Typical of today's lack of social graces, especially on the internet.  The internet allows you to be an asswipe and not suffer the social consequences.
